mScarlet3-S2 achieved up to a 29-fold improvement in stability and is significantly more photostable than any other currently available RFP. Compared to the most photostable RFP, mScarlet-H1, 2, mScarlet3-S2 is up to 4.3-fold more photostable, has a 3.2-fold faster maturation, and exhibits slightly higher cellular brightness (1.1-fold).
During the preparation of our manuscript, a preprint describing a photostability-enhanced single amino acid mutant of mScarlet3, named mYonghong, was submitted to Research Square. mYonghong corresponds to the mScarlet3-S0 variant (mScarlet3-M163H) in our manuscript. We present strong evidence that mScarlet3-S2 is up to 3.7 times more photostable than mScarlet3-S0 under illumination conditions for SIM microscopy.
