miRFP720

similar: iRFP720

miRFP720 is a basic (constitutively fluorescent) near ir fluorescent protein published in 2018, derived from Rhodopseudomonas palustris. It has low acid sensitivity. It requires the cofactor biliverdin for fluorescence.

miRFP720 Sequence

miRFP720 was derived from iRFP720 with the following mutations: S180E/N254V/E301K/L302R/Q305E/V306R/W309T

Note: While the sequence below matches the sequence shown in Fig S1 for miRFP720 in the original paper (Shcherbakova et al., 2018), the mutations from the parental iRFP720 do not. In Shcherbakova (2018), iRFP720 is shown without E180S or V254N, which were described in the text and shown in Fig S1 of the original iRFP720 paper (Shcherbakova et al., 2013). Here, we include S180E/N254V in miRFP720.

Excerpts

Its high extinction coefficient and NIR-shifted excitation and emission spectra make miRFP720, as compared with other miRFPs, an advantageous FRET acceptor for a blue-shifted miRFP670 FRET donor... the red-shifted excitation spectrum of miRFP720 resulted in minimal direct cross-excitation of this FP in a FRET pair.
