To convert mKate into a PAFP, we mutated amino acid residues at positions 69, 84, 148, 165, 167, 181, and 203 (numbering follows the alignment with green fluorescent protein (GFP) , which were crucial for photoactivation of the chromophore in PAmCherry1.
