pH titration showed that mCRISPRed had the lowest pKa value (pKa = 2.1) among all fluorescent proteins known to date, surpassing that of TagBFP2 (pKa = 2.7), those of other comparable long Stokes shift red fluorescent proteins, and that of a green fluorescent protein variant named Gamillus engineered for increased pH resistance (pKa = 3.4). These properties render it very favorable for applications in acidic environments, such as lysosomes.
